Hello.
I want to create a custom alert based on the number of requests.
i.e, I have this service:
I want to have an alert when the number of requests are > 500.
Is that possible? I can not find how to do that. Can you help me?

There is no such option right now. In general if it is unusual DT should alert automaticaly about throughput change. If it is not, open RFE for fixed thresholds for throghouput on services.

And using the Dynatrace Rest Api? I found this (Anomaly Detection - Metric events) but i am not sure.

That's the API endpoint for the "Custom alerts" section of your global Anomaly Detection settings. It is also the endpoint for the Preview of "Custom events for alerting" feature, which will replace that.

I am reading the article. So, i can not create a custom alert based on number of requests using the rest api?

This is possible. You would need to do the following:
Go into your environment settings and select "Anomoly Detecton"
Then select "Custom Alerts"
From there you can select processes and services calls and all the metrics surrounding that call

Yes, like Sebastian said, there is not an option for "number of requests" there.
