When loading the site the below script gets blocked so the site is not loaded

This happens on Chrome browser, also some times? How to fix this?

Most likely this is a browser ad-blocking extension such as adblock or ublock origin.

Without this plugin sometimes the script file is not loaded. Is there any settings on Dynatrace Config?

In this case you have to investigate on a the endpoint where the JavaScript does not get loaded.

Issues may be of various root causes so it's impossible to determinate it without investigating the behaviour in the browser.

Any chance your users are visiting the application via various URLs and the ruxitagent is inserted only on particular ones?

There is nothing you can in the Dynatrace config. I'd only recommend checking the ratio of captured user actions (defaults to 100%), but the ruxitagent JavaScript still should be inserted no matter how the ratio is set.
